Zeiss TIRF III Start Up Procedure.

Startup

  1. Turn on Camera
  2. Turn on Microscope power, SMC, trigger box, stage, definite focus and laser box key
  3. Wait for camera light to stop flashing then turn on the computer
  4. Log in and open Zen Blue Software

Check if the incubation is on, it is usually left on set at 30C. If you want it at a different temperature leave at least 2hrs for the microscope to reach the correct temp and stabilise.

Check the CoolLED light source is off, this is not controlled by the ZEN software and should be turned off for most imaging.

Shutdown

  1. Close software and transfer data to the server or external hard drive
  2. Remove your sample and if you have used an oil objective gently clean the oil from the objective using lens tissue and ethanol, do not use the 70% ethanol or tissue paper these are provided for cleaning work surfaces
  3. Turn off Camera, Microscope power, SMC, trigger box, stage, definite focus and laser box key
  4. Shut down computer
