Zeiss LSM 880 Tutorials for start up, shutdown, imaging parameters and timelaspe and autosave. Video Tutorials Tutorial demonstrating how to start Zen software and how to set up basic imaging settings for Zeiss LSM 880 confocal Basic settings for creating a timelapse in 3D and autosaving the images. Start Up and Shutdown Startup Switch on three buttons on controller (main switch, systems/PC, components) Image Switch on CoolLED, bars on display should go solid black Image Swich on computer* Log in and open ZEN black (not ZEN blue) On the ‘Acquisition’ page ensure ‘Show all tools’ is ticked Find the lasers tab and turn on the lasers, the Argon laser (458nm, 488nm and 514nm) will show red and take a few minutes to warm up. Image In Acquisition Parameters Tab or on the microscope control panel select your objective. Image Put your sample on the microscope, you may need to change the sample holder or use oil depending on what you are imaging and which objective you are using. To focus your sample return to the ‘Locate’ page *At this point the microscope control unit can be used to set the temperature and CO2 levels of the incubation chamber. Do this at least 1hr (preferably 2) before use to allow time for the microscope to get up to temperature. Shutdown Close ZEN software and transfer data to the server or external hard drive (don’t leave Airyscan files hanging around on the server for ages, process them using Imaging Computer 2 and delete the original file) Shutdown computer, turn off CoolLED Remove your sample and if you have used an oil objective gently clean the oil from the objective using lens tissue and ethanol (do not use the 70% ethanol or tissue paper these are provided for cleaning work surfaces) Turn off Systems/PC and Components Wait for fan on lasers to stop (about 5 mins) then turn off main switch.
